Alfa Laval SB Carlsberg Flask is used for sterilizing wort and propagating pure yeast culture for yeast propagation plants in laboratory scale.

Safe, reliable yeast propagation
Developed specifically for aseptic yeast management in the brewery, the Alfa Laval SB Carlsberg Flask sterilizes wort and cultivates pure yeast cultures on a laboratory scale for use in yeast propagation plants in the brewery, dairy, food and beverage industries. The flask can be used as an integral part of an Alfa Laval SCANDI BREW® solution.

Hygienic design
To maintain a sterile environment for safe yeast cultivation, the flask is made of materials that meet stringent sanitary requirements. The design of the flask enables an easy introduction into an autoclave. Recommended sterile air supply during inoculation into yeast propagation plant is 2 bar and must not exceed 5 bar (72.5 psi) at any time.
During transfer of pure yeast culture from the Alfa Laval SB Carlsberg Flask to the yeast propagation plant, it is important that all hose tails are fastened strongly and safely with hose clips.
After use, the flask is taken apart and cleaned manually with normal detergents.

How it works
Construction
The Alfa Laval SB Carlsberg Flask consists of a cylindrical container with a flat bottom and top cover equipped with breathing filters, a membrane sample valve for aeration and product transfer, and micro sample port for aseptic introduction of pure yeast culture.

Operating principle
The flask is filled with wort to the net capacity that corresponds to approximately 80% of the total volume. The sterilization takes place in an autoclave, on a gas burner or an electric hotplate. After sterilization, the flask is placed in a refrigerator or a cold room in order to cool the wort to the desired working temperature. Aeration of the cold wort occurs through the membrane sample valve connected to the aeration lance.
Yeast culture can be introduced aseptically through the membrane fitting by means of a syringe. Alternatively, dry yeast culture can be transferred to the flask through the empty filter housing.
The breathing filter can remain mounted on the flask during sterilization. However, if the breathing filter has been sterilized separately, it is important to remount it at the end phase of the wort sterilization while steam is still evaporating from the flask so that the filter connection is also sterilized. The empty filter housing can be used as funnel for transferring pure dry yeast culture to the flask.
As a second step, aeration of the cold wort takes place through the membrane sample valve connected to the aeration lance. It is important to use sterile air. For safety reasons, passing the wort through a filter mounted in front of the membrane sample valve is also recommended.
Air supply and flow should be moderate in order not to minimize foaming. It is important that foam does not come into contact with the breathing filter because it may lead to blockage and will destroy the filter.
The easiest way to determine the right conditions for air supply as well as duration of aeration (less than 10 minutes) is to conduct tests with the flask lid open. However, yeast culture from these tests should never be used.

When the wort is well aerated, the yeast culture can be introduced aseptically through the membrane fitting by means of a syringe with a content of 150-200 ml.
Alternatively, dry yeast culture can be transferred to the flask through the empty filter housing. In case a larger quantity of yeast is needed, the lid can be disassembled and the culture can be poured into the flask. This procedure, however, requires a sterile environment and the highest degree of precaution must be exercised.
The transfer of pure yeast culture into the propagation plant should take place at the “high kräussend” stage under aseptic conditions. This procedure requires a sterile air supply to the breathing filter. The yeast culture can be pressed from the flask into the inoculator when the membrane sample valve on the lid of the Carlsberg Flask has been connected to a sample device on the receiving vessel (the inoculator).
The connecting hose, membrane sample valve and all other connections must be thoroughly sterilized before this transfer takes place. Sterilization of the membrane sample valve should take place using either alcohol or steam. Between operations, the valve may be sealed with alcohol, which should be blown out with sterile air prior to use.
When the connection has been established and the inoculation is to take place, we recommend opening the sterile air supply first, then the membrane sample valve on top of the flask, and finally the sample device on the inoculator.
When the culture has been transferred (this can be noticed as bubbling noise in the inoculator), we recommend continuing with the air supply for another one to two minutes in order to be sure that all of the yeast culture is transferred.
Finally, the sample device on the receiving vessel and the membrane sample valve on the Alfa Laval SB Carlsberg Flask can be closed, effectively closing offer the air supply.
